A Phase 2 interventional study of Tirzepatide in Sarcoma,Soft Tissue, sponsored by M.D. Anderson Cancer Center. Recruiting at 1 site in United States. Open to participants aged 18 Years to 39 Years. Per ClinicalTrials.gov, last updated 2026-10-20.
Sponsored by M.D. Anderson Cancer Center · Phase 2, Interventional, and Supportive care
This is a prospective, single-arm, open-label feasibility trial conducted at the MD Anderson Sarcoma Center and Cardio-Oncology Clinic. The study will enroll young adults (18-39 years) with soft tissue or bone sarcoma who are overweight (BMI 25-29.9 kg/m² with ≥1 cardiovascular risk factor) or obese (BMI ≥30 kg/m²), have completed Dox-based chemotherapy (≥300 mg/m²) approximately three months prior to enrollment, and have preserved LVEF (≥50%) preenrollment.
Primary Objectives:
To evaluate the feasibility of administering Tirz in overweight or obese AYA sarcoma patients during the first year after anthracycline therapy.
Feasibility will be assessed as a composite of recruitment, retention, and treatment adherence, and will be considered if all the following criteria are achieved:
If any criterion is not met, contributing factors (e.g., ineligibility, refusal, operational barriers) will be documented to guide future trial design.
To evaluate the safety and tolerability of Tirz in AYA sarcoma patients. Safety will be characterized by the incidence, type, and severity of treatment-related adverse events, graded according to CTCAE v6.0.
Secondary Objectives:
To determine the incidence of subclinical cardiotoxicity following Tirz treatment.
Subclinical cardiotoxicity will be defined as the presence of any of the following:
Changes will be assessed upon completion of Tirz treatment compared to baseline.
To evaluate changes in cardiac structure and function associated with Tirz therapy.
Specifically, to assess changes from baseline to after completion of Tirz in:
To assess changes in body composition, dietary habits and physical function/activity.
Specifically, to assess changes after completion of Tirz in:
Physical function metrics including:
To characterize metabolic and cardiac biomarker responses to Tirz.
Specifically, to assess changes from pre- to post-Tirz in:

Time frame: Through study completion; an average of 1 year
Started Oct 2026 (actual). Primary completion is estimated for Dec 2034, 98 months after the start.
Of 211 non-industry phase 2 interventional studies under Sarcoma, half reached primary completion within 67 months of starting (the median), counting those still running; a quarter within 46 and three quarters within 92. This study's planned 98 months is above that.
Counting every sponsor, half of 240 studies reached it within 67 months; a quarter within 41 and three quarters within 92.
