An observational study in Hypertension and Periodontitis, sponsored by Akdeniz University. Completed at 1 site in Turkey (Türkiye). Open to participants aged 18 Years and older, including healthy volunteers. Per ClinicalTrials.gov, last updated 2026-10-14.
Sponsored by Akdeniz University · Observational
This observational study aims to evaluate the relationship between MTHFR gene polymorphisms and periodontal clinical findings in healthy individuals, individuals with hypertension, individuals with periodontitis, and individuals with both hypertension and periodontitis. A total of 200 participants will be included in four groups of 50 participants each. Periodontal clinical measurements, including plaque index, gingival index, bleeding on probing, probing depth, and clinical attachment level, will be recorded. Venous blood samples will be collected for DNA isolation and analysis of MTHFR gene polymorphisms. The study will investigate whether MTHFR gene polymorphisms are associated with periodontal clinical parameters and hypertension.
This is a single-center observational study conducted at the Department of Periodontology, Faculty of Dentistry, Akdeniz University. The study includes four groups: healthy individuals, individuals with hypertension without periodontitis, individuals with periodontitis without hypertension, and individuals with both hypertension and periodontitis. A total of 200 participants will be included, with 50 participants planned for each group.
Periodontal clinical parameters will be assessed using a periodontal probe and will include plaque index, gingival index, bleeding on probing, probing depth, clinical attachment level, and gingival recession. Periodontal status will be classified according to the 2018 EFP/AAP classification. Venous blood samples will be collected from the participants, followed by DNA isolation and molecular analysis of MTHFR gene polymorphisms using polymerase chain reaction (PCR)-based methods.
The study population will consist of adults aged 18 years or older. Participants will be required to be nonsmokers, have no periodontal treatment within the previous six months, and have no systemic disease affecting periodontal health other than hypertension. Individuals with tuberculosis, hepatitis, or AIDS and pregnant individuals will not be included.
The study aims to evaluate the association between MTHFR gene polymorphisms, hypertension, and periodontal clinical parameters. Statistical analyses will be performed using appropriate statistical methods, with statistical significance set at p \< 0.05.

Plaque Index (Silness & Löe)
Dental plaque accumulation assessed using the Silness \& Löe Plaque Index. Scores range from 0 to 3; higher scores indicate greater plaque accumulation
Time frame: At baseline periodontal examination
Gingival Index (Löe & Sillnes)
Gingival inflammation assessed using the Löe \& Silness Gingival Index. Scores range from 0 to 3; higher scores indicate more severe inflammation.
Time frame: At baseline periodontal examination
Bleeding on Probing (BOP)
Percentage of sites showing bleeding within 30 seconds after gentle probing. Reported as percentage of examined sites (%).
Time frame: At baseline periodontal examination
Probing Depth (PD)
Distance from the gingival margin to the base of the periodontal pocket, measured with a periodontal probe at six sites per tooth. Reported as mean value in millimeters (mm).
Time frame: At baseline periodontal examination
Clinical Attachment Level (CAL)
Distance from the cemento-enamel junction to the base of the periodontal pocket, measured at six sites per tooth. Reported as mean value in millimeters (mm).
Time frame: At baseline periodontal examination
