Research objective: To evaluate the efficacy and safety of gematinib combined with lenvatinib in the treatment of advanced or metastatic NSCLC with MET overexpression that has failed standard therapy Experimental design: Prospective, multicenter, single-arm Study population: Patients with locally advanced or metastatic NSCLC who have overexpression of the MET protein (IHC 3+: ≥ 50% of tumor cells show strong positivity; IHC 2+: ≥ 50% of tumor cells show moderate positivity) Sample size: 23 cases (including 6 cases in the safety introduction period) Drug regimen: Gematinib: 300mg, orally, once daily; Lenvatinib: For patients with a body weight of less than 60 kg, 8 mg once daily; for patients with a body weight of 60 kg or more, 12 mg once daily. A 21-day cycle is used until disease progression, occurrence of intolerable adverse reactions, or the subject withdrawing from the study.

Exclusion Criteria:
1. Positive driver genes for non-small cell lung cancer (including: EGFR mutations, ALK fusion positive, ROS1 fusion positive, BRAF V600 mutations, NTRK fusion, MET14 exon skipping mutations, RET fusion); 2. Previously treated with EGFR-TKI; 3. Present with symptomatic brain metastases; 4. Central-type lung squamous cell carcinoma; 5. Currently participating in an interventional clinical study treatment, or having received other study drugs or used study devices within 4 weeks prior to the first administration; 7. Due to gastrointestinal diseases or other diseases that may affect the absorption of the trial drug (such as inability to swallow the drug, severe ulcers, uncontrollable nausea, vomiting, diarrhea, malabsorption syndrome, ulcerative colitis, extensive gastrectomy and small intestine resection, etc.); 8. Having uncontrollable pleural effusion, pericardial effusion or ascites that require repeated drainage; 9. Start is estimated for Sep 2026; primary completion for Aug 2028, 23 months later.
Of 678 non-industry phase 2 interventional studies under Carcinoma, Non-Small-Cell Lung, half reached primary completion within 56 months of starting (the median), counting those still running; a quarter within 36 and three quarters within 81. This study's planned 23 months is below that.
Counting every sponsor, half of 970 studies reached it within 48 months; a quarter within 30 and three quarters within 75.
