Study summary

This phase 1 study will evaluate a single intravenous infusion of BBM-L101, an investigational AAV-based gene therapy designed to deliver a functional SGCA gene, in ambulatory children aged 4 to \<13 years with genetically confirmed SGCA-related limb-girdle muscular dystrophy (LGMD2D/R3). The primary objective is to assess safety and tolerability. The study will also explore muscle protein expression, serum creatine kinase, motor function, biodistribution/vector shedding, and immune responses. Participants will be followed for 52 weeks in the main study and through 5 years after infusion for long-term safety and efficacy.

Read the detailed description

This is a single-center, open-label, sequential dose-exploration study enrolling 6-8 participants. Dose Cohort 1 will receive BBM-L101 at 8.0 x 10\^13 vg/kg as a single intravenous infusion. The dose for Cohort 2 will be selected by the Safety Review Committee (SRC) after review of Cohort 1 safety, tolerability, and preliminary efficacy. Each dose cohort initially includes 3 DLT-evaluable participants; cohort expansion, escalation, de-escalation, or stopping may occur according to protocol-defined rules. The DLT observation window is 12 weeks. Main-study assessments through Week 52 include adverse events, liver laboratory tests, muscle biopsy biomarkers, serum CK, motor-function tests, lower-limb MRI, vector genome/shedding assessments, and immunogenicity.

Participants completing Week 52 enter long-term follow-up through 5 years after infusion.

What these dates mean
Start.
When the first participant enrolled; while estimated, when the study expects to open to recruitment.
Primary completion.
When the last participant was examined or treated to collect data for the study's main question, its primary outcome. A study that stopped early gives the date its data collection stopped.
Completion.
When the last participant's last visit took place, for every outcome, secondary ones and side effects included: the end of the study. Often later than primary completion, sometimes the same day.
Actual and Est.
Actual means the registry says the date has happened. Est. (estimated) is a date the sponsor plans or expects. It can still move, and the sponsor is expected to change it to actual once it has passed.
